Targeted capture and sequencing of gene-sized DNA molecules

December 2016

Authors:
Michael Giolai, Pirita Paajanen, Walter Verweij, Lawrence Percival-Alwyn, et al.

Info:
Scientists adapted the RenSeq capture method for long-read PacBio sequencing, optimizing the method through improved shearing, size selection, and PCR amplification. They used SageELF extensively to remove small fragments from libraries so the sequencer would generate the longest reads possible.

Defining an olfactory receptor function in airway smooth muscle cells

December 2016

Authors:
William H. Aisenberg, Jessie Huang, Wanqu Zhu, Premraj Rajkumar, et al.

Info:
Scientists at Johns Hopkins University, Rutgers, and other institutions teamed up to study receptors in airway smooth muscle cells that appear to play a role in asthma. Results demonstrated that these particular odorant receptors may be a novel target for asthma treatment. Pippin Prep was used to size RNA-seq libraries prior to analysis with the Illumina HiSeq platform.

The mammalian LINC complex regulates genome transcriptional responses to substrate rigidity

December 2016

Authors:
Samer G. Alam, Qiao Zhang, Nripesh Prasad, Yuan Li, Srikar Chamala, et al.

Info:
Researchers from the HudsonAlpha Institute, the University of Florida, and other institutions studied the role of rigidity in the extracellular matrix, finding that the regulation of certain genes relies on nuclear tension. They note that this is the first report of how the linker of nucleoskeleton and cytoskeleton complex alters gene expression throughout the genome. Pippin Prep was used to size-select small RNAs for building a microRNA library.

Selective Y centromere inactivation triggers chromosome shattering in micronuclei and repair by non-homologous end joining

December 2016

Authors:
Peter Ly, Levi S. Teitz, Dong H. Kim, Ofer Shoshani, Helen Skaletsky, Daniele Fachinetti, David C. Page, and Don W. Cleveland

Info:
Scientists report developing a strategy to examine what happens when chromosomes do not segregate properly. They found that missegregation into a micronucleus leads to a cascade of events affecting several cell cycles. PippinHT was used for size selection prior to paired-end sequencing on an Illumina HiSeq 2500 instrument.

Susceptibility to Aminoglycosides and Distribution of aph and aac(3)-XI Genes among Corynebacterium striatum Clinical Isolates

December 2016

Authors:
Jesús Navas, Marta Fernández-Martínez, Carlos Salas, María Eliecer Cano, Luis Martínez-Martínez

Info:
Scientists in Spain analyzed several strains of the pathogen Corynebacterium striatum and found that many were resistant to drugs most commonly used to treat these infections. They used BluePippin to size-select libraries prior to sequencing on an Illumina MiSeq.
